GHTM Sessions 2022 – “Solid-phase isothermal primer elongation using ferrocene-labelled dNTPs for the electrochemical detection of single nucleotide polymorphisms”

  • From: 18/11/2022
  • To: 18/11/2022
  • Location: Sala Fraga de Azevedo | 11:00 - 13:00

The aim of this work was to develop a generic platform for the electrochemical detection of single nucleotide polymorphisms (SNPs) via isothermal solid-phase primer elongation and exploiting ferrocene-labelled 2’-deoxyribonucleoside triphosphates (dNFcTPs). Two examples of the application of this platform will be described, the first using Klenow (exo-) DNA polymerase for the detection of a single SNP linked to resistance to the antibiotic rifampicin in Mycobacterium tuberculosis and the second using isothermal recombinase polymerase amplification to the direct multiplexed detection of SNPs associated with a predisposition to bone facture and osteoporosis.
