The aim of this work was to develop a generic platform for the electrochemical detection of single nucleotide polymorphisms (SNPs) via isothermal solid-phase primer elongation and exploiting ferrocene-labelled 2’-deoxyribonucleoside triphosphates (dNFcTPs). Two examples of the application of this platform will be described, the first using Klenow (exo-) DNA polymerase for the detection of a single SNP linked to resistance to the antibiotic rifampicin in Mycobacterium tuberculosis and the second using isothermal recombinase polymerase amplification to the direct multiplexed detection of SNPs associated with a predisposition to bone facture and osteoporosis.

In May 2020, the Cascais City Council established a partnership with GHTM to conduct a community-level seroprevalence survey, which included blood serology testing for SARS-CoV-2 antibodies together with socio-demographic and clinical questions. The study’s 1st phase was between May and September 2020 and had 12,032 respondents – 5.62% of Cascais population. The reactive tests’ %s were, respectively: 1.3 overall, 1.16 females and 1.56 males; having the 20-39 age group the highest prevalence (2%).

Traditional light-microscopy of stained smears may be replaced by fluorescent stains.
The auramine-O Acid-Fast-Stain (AFS) does not stain the cell-wall (mycolic acids), but is a classical DNA/RNA stain, whereby the cell wall only protects the microorganisms from decolorization by the acid. Confirmation of this new concept opens up the possibility to use this stain in different contexts: a) Auramine-O AFS allows the simple detection of Schistosome eggs; b) Auramine-O AFS allows detection of mycobacteria in histological samples; c) newer nucleic acid-stains may increase (broaden) the application of this stain.

Drones can be used to transport laboratory samples in remote areas, but limited evidence exists on whether that transport affects the samples’ results. Given its potential, the Ministry of Health and the National Institute of Health of Mozambique partnered with VillageReach NGO to generate such data and 300 respiratory samples were transported by two drones, departing from, and returning to the «INS TB Reference Laboratory» in Marracuene district. The samples’ analysis concluded that their quality had not changed by flight, being comparable to land transport.

GHTM Sessions 2022 VBD01 » Optimising strategies for control and elimination of neglected tropical diseases (NTDs)
NTDs cause serious morbidity and disability, affecting 2 billion people in low- and middle-income countries. The main strategy for control of key NTDs is preventive chemotherapy (PC), the regular delivery of drugs to entire communities or specific sub-groups, such as children. PC NTDs of global importance include soil-transmitted helminths, schistosomiasis, scabies, trachoma and yaws. The Kirby’s NTD research group is leading large-scale intervention research linked to novel diagnostics, social science, health economics and mathematical modelling aiming to contribute to the control and elimination of PC NTDs in the Asia-Pacific region and Africa. The RG leader will present an overview of that research with examples including cluster randomised control trials in Vietnam, Solomon Islands and Fiji; and impact assessments of PC programs in Vanuatu, Solomon Islands, Nauru, East Timor and Angola.

There is an overall increase in life expectancy associated to high costs in health care for an ageing population in the industrialised world. Consequently, there is a demand for fast, cheap and easy to use diagnostic tests that are essential in identifying patients, determining prognosis, monitoring treatment and assessing the efficacy of prevention. Here, nanotechnology has a prominent role, with special emphasis on noble metal nanoparticles.
